cialis (tadalafil)

Cialis is the medicine on the basis of tadalafil

cialis (tadalafil)

Pharmacological action:

Cialis is the medicine on the basis of tadalafil. The active component of the medicine reversibly selectively inhibits the cycle guanosine monophosphate (cGMP) that is the specific phosphodiesterase type 5 (PDE5). The local release of NO is registered during the sexual stimulation, and at the same time the specific phosphodiesterase of type 5 activity inhibition leads to the cycle guanosine monophosphate level increase in the corpus cavernosum. Due to tadalafil the smooth muscle tonus decrease and the penile blood inflow increase take place that stimulate erection. Tadalafil does not influence the sexual stimulation. The effect of tadalafil on other phosphodiesterases is insignificant so Cialis does not affect the cardiovascular system, the liver function, skeletal muscle and other organs. Tadalafil does not change the qualitative characteristics of sperm, does not change the level of testosterone, follicle-stimulating and luteinizing hormone in the blood. The effect of the medicine Cialis is possible only under condition of sexual stimulation. The therapeutic effect of Cialis occurs in 15 minutes after administration. Cialis acts within 36 hours after oral administration. The active ingredient of the medicine Cialis is well absorbed after oral administration. Peak of plasma concentration of tadalafil is achieved in 2 hours after administration. Tadalafil bioavailability and speed of absorption do not depend on the food. About 94% of tadalafil is bound to plasma proteins. The active ingredient of the medicine Cialis is metabolized with the help of the enzyme CYP3 A4. Tadalafil is excreted predominantly in the form of metabolites by intestine and kidneys; the half-life is 17.5 hours.

Indication for use:

Cialis is used for the treatment of the erectile dysfunction.

Method of use:

Cialis is intended for oral administration. It is recommended to take whole film-coated tablets with sufficient drinking water or tea. Cialis is taken regardless of food. The physician should prescribe the dosage schedule of Cialis. Usually it is prescribed the intake of 1 film-coated tablet 15-20 minutes before the anticipated sexual activity. Taking into consideration that the effect after a single dose remains up to 36 hours, the tablet may be taken earlier. Maximum recommended daily dose of Cialis is 1 tablet.

Side effects:

While taking the medicine Cialis the patients noted the headache and dyspeptic symptoms. In addition, while taking tadalafil it was reported a few cases of back pain, facial flushing, nasal obstruction, myalgia, pain in eyes, hyperemia of conjunctiva, dizziness and angioedema. There are data on the development of priapism and prolonged erections while taking the medicine Cialis. In the case when an erection persists longer than 4 hours, you should seek medical help as long-term erection may cause damage of tissue of penis and long-term loss of potency.

Contraindications:

Cialis is contraindicated in patients with a known hypersensitivity to tadalafil. The drug Cialis is contraindicated in patients administrating organic nitrates. Tadalafil is not indicated for use by women and children under 18. It is not recommended to take the drug Cialis in patients with diseases of the cardiovascular system, for whom sexual activity is inadvisable, including patients who have recently experienced myocardial infarction or stroke, as well as suffering from cardiac insufficiency of the second degree, unstable angina, uncontrolled arrhythmia or hypertension, severe arterial hypotension. The caution is advised when Cialis is prescribed to patients with an increased risk of priapism, including patients with leukemia, sickle-similar cell anemia, multiple myeloma disease, angular curvature of the penis, cavernous fibrosis and Peyronie's disease. Also the caution should be observed, when Cialis is administrated in patients suffering from disorders of the kidneys and liver, and patients receiving alpha-blockers.

Interaction with other medicines:

Administration of Tadalafil with any form of organic nitrate is contraindicated. Tadalafil does not change the pharmacokinetics of drugs which are metabolized by the system of cytochrome P450. When Cialis is coadministrated with inhibitors CYP3 A4 it is possible the increase the plasma concentrations of tadalafil. In particular, the caution is advised when Tadalafil is coadministrated with ketaconazole, ritonavir, saquinavir, erythromycin and Itraconazole. When Tadafil is coadministrated with inhibitors CYP3 A4, in particular, rifampicin, it is possible the decrease of plasma concentrations of tadalafil. When Cialis is coadministrated with antihypertensive drugs, its action may be potentiated. Antacids and obducing drugs decrease the bioavailability of Tadafil when they are coadministrated.

Overdosage:

When receiving excessive doses of the medicine Cialis the patients noted the development of adverse reactions similar to the side effects of Tadafil while taking therapeutic doses. There is no specific antidote. The standard symptomatic treatment is adopted at the overdosage. Hemodialysis does not significantly decrease plasma concentrations of tadalafil.

Storage conditions:

Cialis expiration date is 2 years from production date in room between 15C and 30C.
